[摘要] 目的:以四物汤古方配伍比例为基础, 考察白芍分别与当归、熟地、川芎不同比例配伍后芍药苷的煎出量,探讨配伍对白芍中芍药苷煎出率的影响,并对四物汤的组方规律进行初步研究。方法:参照四物汤古方中用药配伍比例,利用传统方法煎煮提取芍药苷,用HPLC测定芍药苷的含量。结果:四物汤的组方药物与白芍配伍都能促进芍药苷的溶出,但配伍比例不同促溶程度有明显差异,白芍当归、白芍熟地、白芍川芎的配伍比例分别为1:3、 1:1、3:4时芍药苷煎出率最大,相对于白芍单煎分别提高了48.4%、25.6%、34.8%。结论:当归、熟地、川芎与白芍配伍均能促进芍药苷的溶出。选取的四个四物汤古方中以古方2《石室秘录》卷三方之四物汤的芍药苷溶出率最高。

[Abstract] Object: Based on the compatibility proportion in ancient prescriptions of siwu decoction, decocting rate of peoniflorin in Radix Paeoniae combined with Radix Angelica Sinensis, Radix Rehmanniae Praeparata, Rhizoma chuanxiong by different proportions were calculated respectively. Thus effects of compatibility on decocting rate of peoniflorin in Radix Paeoniae were studied and preliminary study of the herbal prescription rule of siwu decoction was conducted. Method: Referring to the compatibility proportion in ancient prescriptions of siwu decoction, peoniflorin was extracted by water decocting method and the content was determined by HPLC. Result: All the other three members of siwu decoction had positive effects on the dissolution rate of peoniflorin, but there were significance differences between several compatibility rations. Values of the dissolution rate of peoniflorin reached the highest with the following compatibility rations: Radix Paeoniae: Radix Angelica Sinensis(1:3), Radix Paeoniae: Radix Rehmanniae Praeparata (1:1), Radix Paeoniae: Rhizoma chuanxiong (3:4), which were increased 48.4%、25.6%、34.8% than that of singledecoction of Radix Paeoniae, respectively. Conclusion: Radix Angelica Sinensis, Radix Rehmanniae Praeparata, Rhizoma chuanxiong promoted dissolution of peoniflorin in Radix Paeoniae. The highest value came from the second ancient prescription of siwu decoction Shishimilu.

白芍为毛茛科 (Ranunculaceae) 植物芍药 (Paeonia lactiflora Pall)的干燥根,白芍性味苦、酸, 微寒。归肝、脾经, 功效为养血调经、敛阴止汗、柔肝止痛、平抑肝阳[1]。
四物汤最早见于唐代蔺道人的《仙授理伤续断秘方》,用于治疗外伤科疾病,为补血剂的代表方,也是调经的基本方,是后世众多补血方的祖方,临床应用十分广泛[2]。本课题以四物汤古方的配伍比例为基础,研究不同比例配伍对芍药苷的影响。四物汤古方有多种配伍比例以及不同的组方药物,以白芍、当归、川芎、熟地四味药组成的四物汤最为经典。本课题通过对四物汤中白芍分别与组方药物不同比例配伍,测定芍药苷的煎出量,研究川芎、当归、熟地分别与白芍配伍的作用机制,根据临床疗效的不同而确定方剂配伍的最佳比例,并对四物汤的组方规律进行初步研究,从而指导科学配伍,拓宽方剂的临床应用范围。
