植物的种质资源保存技术在当今世界具有重要作用。本文研究了月季种质资源保存技术中离体培养的操作方法。通过培养基浓度的对比,外植体的选择,灭菌材料以及时间的对比,限制生长的植物激素激素浓度的对比,探索月季种质资源保存技术中的最优的种质资源保存方法,并且提出了在月季组织培养中出现的问题及建议,为月季的种质资源保存技术提供实践基础。 结果表明:在月季的发芽阶段,当NAA浓度为0.2mg/l时,选用1.5mg/l的MS+6- *好棒文|www.hbsrm.com +Q: 3 5 1 9 1 6 0 7 2 
BA月季不定芽增殖率最高。MS+6-BA浓度在1.5mg/l~2mg/l时,月季增殖率最高。消毒过程中,75%酒精消毒2min时,月季成活率最高,为78%,2%次氯酸钠消毒6min,月季成活率最高为83%。0.1%升汞消毒4min,月季成活率最高为84%。在月季外植体的选择上,选用中段外植体诱导分化率较基部与梢部高,并且,中段的外植体生长速度最快。在月季生根阶段,选择1/2MS+0.5mg/l NAA+AC效果最佳。在抑制生长阶段,在不同浓度的ABA抑制效果实验对比中,选用1mg/l的ABA,对月季生长抑制效果最佳,而在不同浓度的CCC抑制效果实验对比中,选用40mg/l的CCC对月季生长抑制效果最佳。 关键词 月季,种质资源保存,组织培养,限制生长

Title Study On the Technology of Preservation of Rose Germplasm Abstract Plant germplasm conservation technology plays an important role in todays world. In this paper, the operation method of the in vitro culture of rose germplasm resource preservation was studied. By cultivating the basal concentrations of contrast, selection of explants, sterilization materials and compare the time, limit plant growth hormone concentration of contrast, explore rose germplasm resources preservation technology of optimal germplasm resources preservation methods, and puts forward the problems and suggestions in the tissue culture of Rosa chinensis, rose germplasm resources preservation technology to provide practical basis. The result shows that the rose of the germination stage, when concentration of NAA 0.2mg, choose 1.5mg/l MS + 6-BA rose not shoot proliferation rate was the highest. 月季组织培养,取芽时要在无病虫害的优良品种单株上进行,选取生长健壮的当年生枝条作为外植体,将取回的材料用手术刀片切去叶片及叶柄,切成1-2cm的带芽茎段,将清理好的材料在自来水下冲洗干净,后用洗衣粉浸泡5min,再用流水冲洗10min,洗去泡沫。然后在无菌条件下用75%酒精表面消毒,倒出酒精,无菌水冲洗,然后用2%的次氯酸钠溶液中浸泡,倒出次氯酸钠,无菌水冲洗,最后加入0.1%的升汞溶液灭菌,取出茎段,在无菌水中清洗2-4次。消毒时间进行对比实验,选择合适的消毒方案。在整个灭菌和清洗过程总,应不停地摇动瓶子,使其充分与消毒液接触,否则会影响消毒和灭菌效果,灭菌后的外植体取出后用无菌滤纸吸干水分待用。每种消毒剂接种5瓶,每瓶接种芽5个,以25个芽为样本,选择污染率较低的一种方式。
